Immunohistochemiczna lokalizacja metalotioneiny (MT) w nowotworach nabłonkowych u zwierząt

Metallothionein (MT) was located immunohistochemically in epithelial benign (adenoma) and malignant tumours (adenocarcinoma, carcinoma) taken from different organs of dogs, cats and hairless mice by using the LSAB 2 HRP test (produced by DAKO). The peroxidase activity in relation to 3.3 diaminobenzidine (H202 in 50 mM Tris/HCL, pH 7.8 - NCL - DAB Novocastra) 3 was determined. In addition, the histological preparations were stained with hematoxylin (DAKO Chem Mate). The tests demonstrated that both the MT level and its location are similar in epithelial tumours of different animal species. However, in comparison with MT's stable location in the cytoplasm and/or nucleus of epithelium cells, its content depends on the kind of organ and tumour histogenesis. In comparison to adenoma cells, MT concentration increases in cancer cells. These results suggest that the presence of this protein (MT) may be a sign of the increasing malignancy of animal epithelial tumours.
